Preparing files for iTunes U
iTunes U requires that files prepared for upload are in a specific format. Video must be encoded or exported using H.264 (mpeg-4) compression with one of the following file extensions:

Our recommended tool for converting video and audio for iTunes U is QuickTime Pro. QuickTime Pro is a cross-platform viewer, simple editor and export utility available for a nominal fee (about $30) from Apple. QuickTime Pro upgrades QuickTime player's features and functionality.
