I have long been fascinated with the motion of biomolecules, from the peptide bond to complex machines. In my research lab, we combine biophysical and biochemical techniques to get a close chemical view into biological systems. We explore both disordered and folded protein domains involved in transcription regulation. In particular, we utilize the powerful combination of fluorescence and NMR spectroscopies. Our current research targets three domains within the auxin response factor family of proteins, key to the hormonal growth regulation of a number of food and fuel crops. We will use this system to (1) characterize the intrinsically disordered, glutamine-rich, proteins so common to transcription factors and (2) define the structural basis of specificity in protein:protein interactions or protein:DNA interactions within a set of highly similar proteins.
